(Studies on Modulation of Signal Transduction in Chemoprevention Process and Anti-obesity by Soy-derived Phytochemicals and Related Phytochemicals)

식이중에 존재하는 많은 피도케미칼들은 잠재적인 항암성 및 항돌연변이 능을 보유하고 있는 것으로 알려져 있으며 이들 화합물들의 암예방 작용은 항산화작용 및 항 염증작용 등에 의한 것으로 보고되고 있다. 실제적으로 많은 역학연구나 실험실수준의 연구에서 이들 화합물들은 항종양작용을 보여주고 있고 이들의 작용기전 중 가장 주목을 받은 것은 세포내 신호전달 체계의 변화에 의한 세포증식이나 세포사멸 (어폽토시스; apoptosis)과정에 관여하는 유전자들의 발현을 변화시키는 작용을 들 수 있다. 암이 진행되는 개시, 촉진, 진행의 다단계로 이루어지며 이들 과정 중에서 유전자 발현, 세포주기진행, 어폽토시스 및 세포내의 각종 대사 등을 조절하는 신호 전달체계에 이상을 가져올 수 있다. 세포의 비정상적인 증식이나 악성세포로의 전환 등을 관장하는 여러 신호 전달체계에 대한 도식 (그림 A)에서 보는 것처럼 매우 복잡하고 다양하다.
이들 중 어떠한 경로의 제어에 의하여 세포증식이 억제되고 어폽토시스에 도달하는지를 밝혀내는 것이 연구의 최종목적이라고 할 수 있다.따라서 본 실험실에서는 콩의 이소플라본과 제니스틴의 신호전달체계에 미치는 효과를 조직배양 (cell culture)과 동물실험을 통하여 밝혀내고자 하며 특히 어폽토시스에 관련된 인자들을 포함한 신호전달물질에 미치는 효과를 연구하고 있으며 특히 암예방과 연관이 깊은 것으로 알려지고 있는 Cyclooxygenase-2 및 AMP-activated protein kinase의 조절에 의한 암세포억제기전을 연구하고 있다 (그림 B). 또한 제니스틴은 에스트로겐과 연관성을 가지고 있으므로 에스트로겐과 제티스틴의 효과를 비교 연구하고 있으며 제니스틴이외의 피토케미칼로 녹차성분인 EGCG, 고추의 성분인 capsaicin의 연구도 수행하고 있다. 최근에는 이들 피토케미칼에 의한 항비만, 항당뇨효과의 기전을 신체내의 에너지 균형을 관장하는 AMP kinase의 신호전달 조절측면에서 관찰하고 있다.
